Not sure if openlayers can handle it, but GeoWebCache also supports MBtiles:

Keep in mind that is a community module that needs to be installed.

So you mean to write tiles on disk as a TMS hierarchy. No, that's not
possible at the moment, one would have to write a dedicated blobstore
for it.
