Ciao Arnauld, honestly I am having an hard time trying to understand what you need to do. Please, use less questions marks, don't use uppercase (uppercase == shouting) and give more meaningful details if you want to increase the possibility to get an answer.
That said, It is not clear to me whether or not your tiles on disk are simple png files without georeferencing or if they are georeferenced raster tiles (e.g geotiff). Moreover, are we talking about a mosaic (i.e. large number of relatively small tiles creating a tessellation of a certain area) or a pyramid (same thing but with multiple resolution levels).
