If you're using the deprecated (AJAX) Search APIs (e.g., Web, News, Images), you don't have to sign up anywhere, per se. (I would not, however, recommend doing so.) However, if you're trying to use the Custom Search API or any of the newer APIs, you will need to use the APIs Console to obtain an API key and, particularly in the case of a web application, set up access controls. You can do this at http://code.google.com/apis/console .
