For large numbers of markers try Bill Chadwicks "Block Markers", available at http://www.bdcc.co.uk/Gmaps/BdccGmapBits.htm It might be worthwhile doing some measurements of (a) load time and (b) page refresh time for certain numbers of markers, using GMarker and Block Marker.
