Hi Amol, The map usage (or map load / page view) is only counted each time you load the javascript API. The moment the javascript API is loaded, this counts as a deduction in the map load quota. Within that map load, you can perform directions look-up, geocoding, distance matrix, elevation, etc and it will still be considered the same map load.
If you're using the web service APIs for directions, than each request to the web service will count as a deduction in your daily quota for that server (i.e. for Free maps users, the directions web service has a daily quota of 2500 directions requests, so each request will deduct 1 from your daily quota bucket).
