On my system, launching the app again from the Apps folder or Spotlight 
brings up the prefs pane. Try quitting the app from the Activity Monitor, 
then launch it fresh and see if that brings up the prefs pane.

Also, from the Growl notification that pops up when selecting No Icon:

*Enabling this option will cause Growl to run without showing a dock icon 
or a menu item.*
*

*To access preferences, tap Growl in Launchpad, or open Growl in Finder.*
*
*
... so it should work.
