I'm seeing something similar after the update. If I click the GrowlTunes icon, I see what is currently playing, but am not getting notifications.
I do have preferences selected in GrowlTunes for everything. Growl 2.1.2 is otherwise working with everything else. I disabled the iTunes built-in notification center notifications.
