The main tools I've seen real projects done in are:
* HAppS (see hpaste.org)
* fastcgi + xhtml + maybe some JSON (used for lots of things)
* cgi + xhtml
* WASH (see braintreehemp.com.au)
So it depends on what you're comfortable with.
