I'm wondering whether there are any recommendations for how to effectively cryosection dorsal mouse skin for immunohistochemistry. I'm having difficulty with much of the mouse skin slipping off the slide during the fixation and washing steps, which seems to result in loss of the epidermis in particular, that part of the tissue that I happen to be interested in. I apply the dorsal mouse skin cryosections onto SuperFrost Plus (Menzel-Glaser) slides and air dry them for 2 hours. I use acetone for fixation, 10 minutes, and use PBS for washing. Any suggestions?
