I am going to stain rats' brain microvessels with ER(estrogen receptor) antibody. We know how we can isolate the vessels but this is the first time to stain them. I have the microvessels in PBS 1X and do you know how I can fix them with paraformaldehyde and mount them on the gel-subbed slides? I did 2 ways: 1- mount the fresh microvessels on the slide and wait overnight to get dried and then fix them in 2% PFA for 30 min and after that washing steps and the rest of IHC protocol or 2- fix the vessels with 2% PFA in the tube for 30 min and then mount them on the slides and wait overnight to get dried. I am hoping that someone out there can lend me some assistance. We currently are using paper slide labels( 0.875 in by 0.875 in) on our Pathology slides, an old label printer (Genicom Model 1025) and Meditech Magic software. Our dilemma is this... we no longer can find the ink ribbons for our printer. Is there anyone out there who is using the same system? In about 1 1/2 to 2 years, we are going to be changing to a new software (possibly McKesson), so we are not looking to really buy anything expensive at this time. We just really need to continue using the paper labels until we have the change over. Thanks for your help.
