If you are using an H&E it was vertually impossible in our hands to try to 
threshold fiber area.  We worked with Image Pro Plus for about 6 months trying 
to get this to work and we could not, we ended up creating an algorithm in 
image pro that allowed us to trace the fibers and that would calculate fiber 
area and diameter, etc.

Subject: [Histonet] ImageJ : start help for muscle fiber count

Any guru out there that might give me some ideas how to measure size and number 
of muscle fibers in a cross section using ImageJ
Area by using freehand tool  works................. but I'm having problems 
making a threshold with the muscle cells outlined for automatic measurements.
 I'm not experienced, might be I haven't found the correct tool :-) Picture
