Does anyone know how I can edit an ASCII file found in a directory,
using the ISHELL from inside of TSO and the E command next to the file I

If your on a current release of z/OS, why not use 3.17?
Get a directory list, place the cursor next to the filename
and enter 'ea' as your line command (Edit ASCII)?
