Just to give you my experience:

I have approx. 4000 users under one of my domains, am using an exteral DB
(MS SQL), and I experience the same thing, only not quite so long (about 30
seconds).  When you expand the users tree, Imail administrator basically has
to query the database for each user (so opening your users folder initiates
~80,000 database querys).

While inconvenient, this has been about the only operational flaw I've
experienced in regards to the amount of users.  We administer our users thru
our billing system's interface into SQL, so the only time I even open the
administrator interface is to add/edit/remove a mail host (we host email for
approx. 200 domains), or to inspect the mail queue, logs, etc.
