Yes. If you have IP addresses, or blocks, that you want to block, that is what it is there for.

using the Control Access feature in this manner? I forget the actual file name, but you do not edit the file. (Although Ipswitch has it in the KB on how to do it.) You control it through SMTP settings, Control Access.
