3 options:

1) The easiest thing is to hold down your left ctrl key, and right-click
   in the xt5250 window.  You'll get a list of different sizes to choose 
   from.

2) If you want this to run every time you start up xt5250, you can change
   your xt5250 script.   It should now be installed in
   /usr/local/bin/xt5250.

   Change the line that says:
   exec xterm -name "xt5250" bg black -fg white -T "xt5250 -$XT5250_HOST" \

   to say something like (adjust to your liking):
   exec xterm -name "xt5250" -font 10x20 -bg black -fg white -T "xt5250 -$XT5250_HOST" 
\

3) If you're running the current development sources, you can set your
    font size in the tn5250 config file by setting font_80 = 10x20,
    font_132= 10x20
