I prefer to use the 1st/6th courses as a tonic unless I have to play 
with other instruments, then I use C, which amounts to the same thing 
except 2 cents flater.

It's more a question of what notes work together. Like Stewart just 
mentioned in the context of choosing certain chord shapes: G# vs Ab; 
F# vs Gb. All the regular lute keys sound great.

I use a Peterson VSAM tuner which has a couple of user programer 
presets, but I used a regular tuner for years. Without a tuner, or 
with a regular tuner, I start with getting my 1st and 6th in tune and 
then I just try to get the 2nd course a couple of cents flat, the 
third 4 cents flat, the 5th 2 cents sharp, the 4th 4 cents sharp. 
Easier said than done on a lute.
