I assume you're talking about a "poster paper". The answer is "Yes, LyX can be used for posters: I've done it." I don't recommend it. I've also tried OpenOffice, and I certainly don't recommend that, either. What worked best for me is Scribus. I've also found that it is best to work with a smaller format than the final poster. My last poster paper required panels 60 inches by 35 inches; I prepared them as 20 inches by 11.67 inches, and scaled them to 300% when sending them to the plotter.
