Multiple signatures for your different email accounts under iOS 5 are still not 
supported, but I can suggest two different work-arounds.

First, iOS 5 supports the creation of keyboard shortcuts.  If you go to 
Settings > General > Keyboard there's a heading for "Shortcuts".  The first 
entry under "Shortcuts" gives an example: "omw" for "On my way!"  If you double 
tap on this sample entry, you'll view the detailed shortcut screen for the 
example, with text fields for the phrase "On my way!" and for the shortcut 
assignment "omw".  There's also a description "Create a shortcut that will 
automatically expand into the word or phrase as you type".  You can make any 
editing changes in the text fields for the shortcut and phrase, then double tap 
the "Save" button in the top right corner.  Or, you can double tap the 
"Keyboard, back button" at the top left corner of the screen to return to the 
keyboard settings screen.  

Just after the sample "omw" shortcut is the "Add New Shortcut" button, which is 
also the last element on the screen.  So you can always navigate to it with a 
four finger tap on the bottom half of the screen.  Double tap on this button, 
then type in the phrase you want to shortcut in the text field.  Then flick 
right when you're done to type in the text for your shortcut abbreviation.  
Double tap the "Save" button in the top right corner.

So the way this could work is that you might  set up your basic signature in 
Settings  > Mail, Contacts, Calendars > Signature (where the default says "Sent 
from my iPhone"), and then customize each additional line you'd like to add by 
typing your shortcut.

The main drawback of this method is that you still have to type one shortcut 
per line of expanded text.

The other option that I use is to use TextExpander Touch ($4.99 by 
SmileOnMyMac) to expand text.  You can define multi-line snippets in this app, 
and download the basic predefined snippet definitions.  Then, if you type 
"ddate" ("date" with an added "d" at the beginning), it will expand to the 
current date, and in the format that you set up.  You could have a multiline 
signature that you define as "ssig1" (starting with double letters for you 
snippet shortcut definitions is good for avoiding confusion with real words).  
This lets you customize everything.  The main drawback is that iOS apps like 
Mail are sandboxed, so you would have to double tap the button to copy to 
clipboard, then use the App Switcher to switch to mail, and then paste.  Or, 
you could compose mail in TextExpander and send it out directly from there, but 
that doesn't let you read through your mail and reply.

I'll just note that TextExpander's snippets for HTML and CSS programming are 
really clever, and that most good notes, Twitter, and messenging apps will 
support TextExpander so you can compose using your custom-defined snippets 
within those apps.  As for HTML coding, the control sequences will expand and 
leave your insertion point between the control elements if you use the 
TextExpander HTML snippets.  So you don't have to type out starting and closing 
elements by hand.
