If Sender: is missing, the application will most likely use From: for envelope sender (MAIL FROM:) (normal MUAs do that). That will make SPF fail and bounces will be send to the original sender of the message instead of to the mailing list system (so it can unsubscribe them). Sympa might handle envelope from different, but please try it out.
