If Sender: is missing, the application will most likely use From: for
envelope sender (MAIL FROM:) (normal MUAs do that). That will make SPF
fail and bounces will be send to the original sender of the message
instead of to the mailing list system (so it can unsubscribe them).
Sympa might handle envelope from different, but please try it out.
IIRC sympa directly invokes the sendmail command, so sender can be set
by the -f flag and you need to make sure it uses your domain for MAIL
FROM.
