I don't think that it's possible to automatically pick up multiple directories in one pass. You can make an XML version of the RIC using the utility posted on this page: https://trac.osgeo.org/fdo/wiki/FdoGdalNotes#CreatingTileIndexConfigFiles The code only looks at a single directory though; you will need to it to recurse into subdirectories if you need it to do that.
