The Mapnik portfile has gone unmaintained as I've focused energy on the binary packages available at http://dbsgeo.com/downloads.

Even if the portfile were maintained i dont recommend macports as their installation approach for boost is error prone. But in this case, essentially the problem with the port is that boost was upgraded by macports and the pymapnik portfile still uses Mapnik 0.6.x. So you should file a ticket at macports.org to upgrade the mapnik portfile.
