I like this answer on Amazon better, if it's still sealed don't mess with it, if it got air in it pull a vacuum to get rid of the air. Air is not a refrigerant, at the very least it's an impediment to refrigerant flow:

A:
Only if you have a leak and your system is empty. Then you will need to pull a vacuum on it after you have repaired the leak. Other wise you can put a can of stop leak in it and then change it back to the normal amount needed. Hopes this answers your question.
