Hi,
the Nikon NEF file format is not supported by MITK. TIFF (16 bit) is
currently also not supported but may be an interesting feature (see
feature request http://bugs.mitk.org/show_bug.cgi?id=10823).
At the moment you can convert your images to a file format which is
supported by MITK (perhaps png is a good idea since it also supports 16
bit) and then load them into MITK.

Seems that 16 bit or float tiffs are not supported. NEF allows loading
data but shrinks the image to a very small size (100x160 pixels from
4288x2848), also the bit depth gets downscaled to 8 bit data. Also big
data in 8 bit tiff mode gets prohibited (not possible to load).
