It is recommended, but not required. At the time that SCOM 2012 R2 shipped, and the upgrade testing was done, the current shipping UR for SCOM 2012 SP1 was UR4, which shipped at the same time as SCOM 2012 R2.
UR6 contains some very specialized fixes that are not included in SCOM 2012 R2. In order to have the "safest" upgrade, I'd recommend always applying the UR that was most tested when the upgrade process was being developed. In this case, it would be UR4 for SCOM 2012 SP1. In your case, I'd recommend simply applying UR4 to your SCOM infrastructure, and skip any agent based updates. Then when this is stable, upgrading to SCOM 2012 R2. When that upgrade is deemed stable, apply UR2 for SCOM 2012 R2. Or, just try the upgrade from your existing SCOM 2012 SP1 UR2 level. The documented Microsoft "recommended" approach will always assume you are on the latest UR before a major version upgrade.
