Not necessarily; as long as the clients can access the SUP on the primary you would be ok. If the connection from the remote site is unreliable, or extremely slow, that might be a problem if you have a large client population at the remote location. If you deploy the update packages to that remote "DP" then the clients would use the packages deployed to that server, and access the SUP from the primary.
