You cannot target Task Sequence advertisements to specific servers. The only way to do what you're looking to do is to scale back the original advertisement from all system to a collection of all systems minus the new site. Then create a separate advertisement for a collection corresponding to the new site. Two advertisements to two mutually exclusive collections = no overlap.
