Doesn't that need to have a trailing / if it's a Maildir and not
an mbox file?
set spoolfile=~/Maildir/

MAIL is a shell variable, your mail spool, and is usually
something like /var/(spool/)?mail/$USER. It is ! in .muttrc's.
