Maybe, an automatic transmission is a complex hydraulic system. ATF is designed to keep it clean and sealed for essentially the car's lifetime. so it is a lot of cleaners, and seal conditioners mixed into a quality hydraulic fluid. If your issue is leaking from a hardened o-ring then it might work. It won't fix a leaky head gasket.
