Although your "start on startup" can be changed to something else
as pointed already, your script might have started and failed. You
can check by changing your script to:

script
    export HOME="/root"

    logger "My app starting..."
    /usr/local/bin/node /root/test.js 2>&1 >> /vag/log/node.log
end script

Reboot and check your syslog.
