You can set “location=doublesided” but “duplex” isn’t a valid argument and even 
then it works only when you have a double sided document.

\setuppapersize[A5][A4]
\setuplayout[location=doublesided]
\setuppagenumbering[alternative=doublesided]
\starttext
\dorecurse{20}{
\input knuth
}
\stoptext
