You probably need the opentype features activated. Ezra has two features from what I can tell, so try something like this (totally untested):

\definefontfeature
   [silezra]
   [mode=node,language=dflt,script=hbr,
    mark=yes,ccmp=yes]

\definedfont[EzraSIL*silezra at 14pt]לָכֵן
