No, sorry.

I need to stabilise a close up a closed eye with makeup being applied. I
can use planar tracking to get perfect tracks of regions of skin, and
then use this to get splines to perfectly follow contours of the eye. If
only I could lock the destination splines and have the source splines
track I could stabilise! However I can only apply the track to both the
splines. I can do it if I animate the spline by hand but this seems an
unnecessarily time consuming way to do it.

If you have roto splines that follow the shape, you should be able to set the source curve to be expression linked to the tracked shape to help you achieve this. The destination curve can be manipulated separately from that.
