You can create it using MAKEDEV. if however, it cant be created by MAKEDEV due to any reason, you can use mknod to create ttyS0.
mknod /dev/ttyS0 c 4 64 chown root.root /dev/ttyS0 chmod 0644 /dev/ttyS0
Note: on some distros, ttyS0 belongs to dialout group.
