you have to add that line yourself, if you decide to go that route.  AFAIK
maya will not auto-add anything to your Maya.env

yes.  i'm curious what would make you think that it would not since that's
what the System Env installation instructions say to do?

if you carefully follow the manual installation instructions for both 2010
and 2011, and point both versions to the same pymel directory on your
system, you should not have any problems.

the only caveat i am aware of is this issue posted about Vista:

no, the python included with maya is all you need. once again, the
instructions would tell you if you needed to install python.

i'm sorry to be a little bit rude, but please RTFM.  i spent a lot of time
writing it. it's not perfect and things are always changing --- new OS's,
versions of maya, etc -- but barring anything unforeseen it should get you
all the way to a working pymel. if there are parts of it that you feel
should be improved, please make a ticket like the one i posted above and
we'll fix it up ASAP.  if you run into problems once you've given it your
best shot, post your problems here and we'll get you sorted out.
