Yes and no, big todo qmail can not read a "little" todo qmail queue.
No because only todo messages are affected (at least afaik).

The normal update from little to big todo is:

1) install a big todo qmail in a different temporary location.
2) start the temp qmail (now both, little and temp qmail are running (only
qmail-start)
3) switch the smtp from little qmail to the temp qmail (this includes all
other mail injecting apps (qmqpd, qmtpd and sendmail))
4) wait until the queue in little qmail is empty (takes max a week)
5) remove little qmail and replace it with a big qmail
6) run big qmail (togehter with temp qmail)
7) switch all injecting processes (smtp, etc) to big qmail
8) wait until temp qmail has no mail in queue
9) remove temp qmail
10) your system is updated.

An other sollution is to use a big todo capable queue fix tool but that's
not so straight forward.

There is probably a possibility to do it without all this. If you stop the
injecting processes then check that all mails are preprocessed, it should
be possible to update in one step but it may be possible that you loose
some mail. So this is only for admins searching a thrill (as the sunsrise
mail admins here in switzerland who trashed all emails of 500'000
customers).
