The latest development version of QmailAdmin contains this feature, but requires vpopmail 5.3.20 or newer. Note that the development versions haven't been fully tested, and probably still have some bugs.

To manually set the Reply-To, add an entry to the "headeradd" file in the mailing list's directory (typically ~vpopmail/domains/domain.com/listname).

I just realized that you also need to add "reply-to" to the headerremove file, something that QmailAdmin 1.0.27 currently does not do.
