I will not recommend it.

You might get lucky and it will execute without any problems, but, I
personally, will not risk it (due to my past experiences).

I batch my structure changes.  So at the end of the day (when I'm the only
one left in the office aside from the janitor), I connect to the database in
single user mode and make the appropriate changes (after a back up and
unload file has been created).  "Better to be safe than sorry."

while users are using the system?
