Hello,
I read somewhere that Heng Li said that bam files should be smaller after
sorting because sorting gives a better compression ratio. However, for me,
after sorting my bam file went from ~31 GB to to ~34 GB. I was wondering if
this is normal or that there is something wrong with my bam file? For
conversion, I used sambamba, and the samtools for sorting, would that have
made any difference? I was under the impression that it wouldn't because
sambamba was only meant to be faster than samtools.
The command I used for sorting is:
samtools sort -m 4G -@ 8 -O bam -o CB2_sorted.bam -T /scratch/tmp/
CB2_header.bam
