I am a graduate student at the University of Vermont working on a mycorrhizal ecology project. I have encountered an interesting 'problem' that I am currently trying to resolve and I would appreciate any input.

I have been staining roots of grasses to quantify infection status. I have used two stains: ink and trypan blue. They provide very different results. Does anyone know if ink preferentially binds to mycorrhizal structures whereas trypan blue is a more general stain, picking up all fungal structures or if trypan blue is a better overall stain picking up mycorrhizal structures that ink misses? If so, I would appreciate any input and pointers to the literature on the efficacy and specificity of these two stains.
