Checking off System State as well as information stores covers it.  I do a
normal file level backup in addition to this but don't include the
Exchange databases since the Exchange Agent takes care of these.

So basically on each E2K server you need to backup the stores and the system
state, and then you're covered? (And of course the system state on the DCs
too.)

Same here stores and system state. I just do a file level backup once week
or after some change.
