No, you just rebuild from source.  .desktop.in files will be converted
to .desktop files which includes your translation from a PO file (that's
what intltool does for you :).


If you really care about how it's done, you can do it manually:

  intltool-merge -d ./po ./game1/game1.desktop.in ./game1/game1.desktop

(and you'll have to repeat this for every .desktop.in file if you
don't want to rebuild from source, and then install resulting .desktop
files where appropriate, usually /usr/share/applications)

Those translations are kept in .desktop files, i.e. outside of .mo
files.  They are installed when you make install a package, so
you'll need to recompile from source, or wait until your distributor
includes your translation. 

The same will happen with many other files, such as .xml files
(eg. for locations in gweather), .schemas (as seen in gconf-editor),
etc.
