Good morning Histo folks, I am hoping somebody can help me. Today was a horrid microtomy day, the static was brutal. I tried putting lotion on my hands, the fabric softener sheet, and wearing gloves but to no avail. I heard rumors somebody used to manufactor an anti-static brush but have yet to find one. Does anybody have any other tricks that could be shared? I am not sure where you live so this may or may not apply. I have lived in several placed and one I learned no matter where I am when they turn on the heat and you wear anything with polyester or that can become electrostatic we had issues with static. I am not sure how to fit it as I have made sure I touch something metal to discharge the static. It does not cure the problem only humidity will and being careful not to start to cut fully charged so to say. I used to have one of those anti-static brushes, it had a small radioactive piece of metal that discharged the static. It worked sometimes. Try putting moistened gauze, or the kimwipes used to clean your waterbath in your microtome wax waste tray. That seems to help me. Good luck. Before I start cutting I spray StaticGard on the arms of my lab coat. I don't know what lotion you use but a non lanolin based moisturizer helped me (corn huskers lotion - it is glycerin based). This works for some but it comes down to trying different things to see what will work for you. To all you wonderful folks who responded, a big thanks! I've been doing histo since 1990 and NEVER had static like today, it's not usually that big of a problem in this mid-Atlantic state. When it does occur the tricks I named earlier usually help but I am always up for learning something new. I will try the low tech option of moist something in my clean-up tray tomorrow, first. I usually can't stand anything in my tray after facing but if given a choice I'll suck it up. I will keep you posted. Thanks again! Product no: 51922 5.7cm Handle length www.tedpella.com it works very well, I clean the metal surfaces of the paraffin microtome and do not wet the brush. Just be sure to spray the floor and the soles of your shoes, as well as the microtome and benchtop. Also, some microtomes have a grounding strap or a place to attach a grounding strap. Or you can wear a ground strap on your wrist, although this is a little restrictive. The best solution IMHO, ground your microtome and use professional anti-static spray (Not from the grocery store.) Thank you I should give more details -I am not actually doing histology but trying to find a method to preserve tiny plant shoot apical meristems (approximately 0.05mm cubed in volume) so I can then measure the amount of light organic elements (CONPS) in a section by using x-ray dispersive spectroscopy with scanning electron microscopy. I thought a variant of Karnovskys fixative with no buffer may do it-mix equal amounts of formaldehyde (say 16%% pure with no methanol and not 10% buffered formalin) and glutaraldehyde (25% EM grade)-so the formaldehyde penetrates the tiny sample very quickly with fast initial fixation and then glutaraldehyde gets in more slowly but gives more extensive stronger fixation. Am I wrong-will the formaldehyde initially harden the sample so the glutaraldehyde may not get in ?. Should I use the lowest formaldehyde concentration that will give initial fixation ( say 8% which will end up as 4% when diluted with equal amounts of glut.). Please let me know what you think. It is a real issue as we are basing the 10% number on the amount of formaldehyde (37 - 40%) added to buffer or water not the actual amounts. Sorry it is confusing about amounts.
