[Histonet] Multiplex IF staining
Hello Hist-world, Does anyone have experience doing FISH/double IF staining? My newest project is to combine a FISH probe (texas red) with anti-Staufen IF (green secondary) and anti-MBNL1 IF (secondary TBD). So I've got a red, green, and blue (DAPI counterstain) already. If I used a yellow secondary for the MBNL1, would you be able to see the difference between that and the green? I'm running out of colors here! Any help would be greatly appreciated. [Histonet] Fixation for FISH
Hello Histo-land, I'm going to start doing FISH in our lab, but I've never done it before. I read that the PFA fixative solution should be made fresh; however, I have some PFA that is being stored in the -80 freezer (for less than a year). Does anyone know if this would be ok to use? And actually, if it is ok, I thought about making a batch of 3% PFA and aliquoting it into smaller containers to freeze and to use at a later time. The only reason I thought about doing that is that I've heard to make it can take awhile. But this is all new to me, so any and all help would be much appreciated. I might as well add this as well: specifically, I'll be trying to optimize a protocol for Texas red-2-O-methyl-CAG probe with IF for the MBNL1 protein. I've read that combining these 2 methods can be trickyand I've never done either of them! Is there anyone out there that can give me some advice? We haven’t changed our mounting medium, but I’ll have to check on the alcohol levels. When I stain (we hand stain only - very small lab), I always make sure the alcohol is higher than the eosin, but I have a lab assistant now, and it’s very possible I didn’t make that a point. Their emails are included on this post, so I'm going to turn it over to them:) But here is their description of what they're doing: In answer to Jack's questions (I thought about replying myself, but didn't want to confuse anyone), we want to make a CEMA array (cutting edge matrix assembly array), which could be done with a variety of tissue types. The step that calls for the special microtome is when you cut the original blocks from the hospital -- meaning variable tissue depth. Though the article mentioned a microtome that could cut 50-2000, we are most interested in the range around 50-150 microns. Hello, I know this is probably a long shot, but I'm trying to set up a stainer and don't have a user manual. It was bought at auction by my company, so who knows how old it is! It is a Microm DS 50, made by Zeiss (now Thermo Richard Allen). I've been trying to get my hands on a manual (having trouble setting up a staining program), but I don't quite know if I'm going about it the right way...or if there is one I can actually get.
