formaldehyde. We get this from Fisher and the number is HC426043.

We use acetone to denature fatty tumors without any problems. They sit in it 
for at least half an hour. I don't rinse them.

Acetone doesn't belong in the grossing area - too much of a fire and explosion 
hazard. If your pathologists are using it to fix marking ink onto specimens, 
they can use 3% acetic acid (half-strength white vinegar). I don't use anything 
at all - if you blot the specimen dry before you put the ink on, it'll stay put.

Another +1 for the VIP E300 (very similar model).  We have one that's close to 
20 years old that's still running.  It has an annual PM and any repairs have

Does anyone have input on the Sukura E150 tissue processor? Our research lab is 
looking at purchasing a used model from Rankin Biomedical, but we were

My hydroquonine was received in the lab in 1980 and it works.  As long as my

containers to do the rest of the stain by hand?

[Histonet] antigen retrieval question

If anyone out there is using a decloaker for antigen retrieval, when doing ihc 
staining, would you please contact me on my e-mail address? I have been 
struggling with a BVD test for months. It is not working consistently and I

[Histonet] RE: Sakura film coverslipper

We have had a Sakura film coverslipper since 1997. It has had three service 
calls, mostly in last 5 years. Those service calls were minor problems quickly 
fixed. When we demoed several different coverslippers, we found this one to 
have the least amount of tweeking involved. It has been a wonderful machine 
and Tech One services it. They say it is a great machine. If I were buying 
again; I would go straight to this.

Both animal disease labs in the state of Illinois have used the Sakura machines 
for the twenty years I have been here with great success. We also have aSakura 
processor that has been going for 25 years and a coverslipper that has only had

I am also in the market for a new paraffin embedding center.
I have demo-ed or site-visited the Sakura TEK5, the Leica EG1150, and the 
Thermo-Fisher HistoStar.
I was wondering if people could give me their critical opinion on these, or 
other machines.
What sorts of problems have you had with them.

I currently have a 25 year old Shandon Embedding Center. I like it a lot.
But I would like to find a machine with a specimen holding tank large enough to 
allow me to immerse 300 cassettes all at once.
This is because I infiltrate brain tissue with Tissue Path Paraplast but embed 
with Surgipath Embedding Media So I let the cassettes sit immersed in the 
Surgipath  for an hour or two before embedding.

(Until I can buy a new processor, The Shandon's holding tank also serves as a 
third processing station, since my Shandon Hypercenter has only 2 wax 
reservoirs) I also do not feel comfortable having the cassettes sitting dry in 
the holding tank

Hmmm...I think more people should have paid attention to Obamacare two years

While this stinks on many levels, I have to take issue with the shift wealth 
from specialists to family practice - family practice docs have been the 
frontline of medicine, all the while earning less than a quarter of what 
specialists earn. It's about time they get a boost.
Too many specialists earn over a million a year, while the family practice 
guys/ladies can barely pay their student loans.

We had Ventana and changed to Dako.  The Dako phone tech support is
excellent and we never wait long for in house service.   Our

We work in a previously underutilized histology lab.

There was a sliding horizontal Sakura microtome, a functioning Thermo 
microtome,
and a nonfunctioning Leica 5xxx (not a very fancy Leica).

The center where we work has no interest in fixing the Leica microtome.
I am supposing that it is merely dirty.  Beijing is very dusty and the 
windows to
the room leaked badly until we installed window seals.

I would like to try to take the Leica microtome apart and try to clean 
it but I cant find
the way to get it open.  There is no hatch to open like on an old AO 
Spencer.

We are told that it would cost way to much to have Leica service it.

Does anyone have any experience with maintenance or repair of a low-end 
Leica Microtome
and have any tips to share on how to open and clean it?

We have used a mixture of Eosin Y and Phloxine B with acetic acid as follows:
780mL 95 %ETOH
100mL Eosin
10mL Phloxine B
4mL glacial acetic acid
We seem to be consistent. We have used this for 17 years.

Hi Folks...I am hoping you all give me a little help.  Our Pathologists are 
complaining about our Eosin on the HE's being weak. The funny thing is, is 
that it can go one

day to the next...one day it looks great...the next it is weak!!

I have already done some experimenting with...1) time tissue spends in 
Eosin 2) making sure that the  alcohols after Eosin are the proper 
concentrations3)

 reducing the time that the tissues spends in the alcohols atfer Eosin... I 
have even gone as far as 4) increasing the rinse time in water after the 
decolorizing and bluing

 steps. 5) I have checked the pH of the water as well.

Any help and suggestions would greatly appreciated!!

I, personally, found the joke very funny. I find a lot of distasteful jokes 
funny - I actually prefer them over anything clean. That does not take away the 
fact that discussing religion, or politics (with humor or in any other
form) has no place in the workplace. Histonet is, in many ways, an extension of 
the workplace. I also do not discuss religion or politics with strangers, and 
there certainly are more strangers that read this blog than folks we know. 
While I was not personally offended by that joke, it is very conceivable to 
think that some folks would be offended.

As I told one replier - had this joke been about Jews it would have been 
something folks reacted to harshly. And, for good reason. So we cannot joke 
about Jews or Muslims, but Catholics are fine? I respectfully disagree - ALL 
religions and posts of humor in reference to a religion on a public listserv is 
a terrible idea.

And, incidentally, this support for those that could be offended is coming from 
me - a person that thinks ALL religion is a joke in the first place.

The ultra view platform changed its clearing process in November. After 
staining slides are to be run through two changes of soapy water and then 
distilled water. After that is done they go on a slide dryer/warmer for at 
least 15 minutes before going into xylene and then coverslipped. Our sales rep 
made me aware of the change when it happened. I opened a new kit yesterday and 
what I just wrote is what the package insert said to do.

One great thing is that it does everything on the machine. It deparrifinizes as 
well as stains. The ultra view platform has even simplified the steps from 
machine to coverslipping. It can be set to run overnight. It has delayed timers 
for starting or ending.

We have used this product for many years as well. It has performed well without 
the hazards of xylene. We still use xylene on the processor for cleaning. I 
have not used it in special stains, only on the processor.

I have used this product for about 10yrs and love it.  It has no fumes and
no foul odor like the orange smelly stuff which gives me the worst
headache. I use it in my processor. To deparrafinize and in my routine
stain line. It works great with special stains to. I have not used it for
IHC since the lab I work in does not deal with that test. It is non toxic
as well.

I purchase my supply from Chad at Mercedes Medical.

P.S. There are few draw backs. I feel that the solution has to be changed
more often than xylene as it becomes saturated faster. It also does not
remove coverslip as well as xylene.

I find it amazing sometimes when you don't do something for awhile how quickly 
your brain throws the information away.  That being said...I know back in the 
day when I was learning histology we used to make our own acid alcohol solution 
(now where I am had a butt load of Clearifier so I was using that up).  I don't 
want to buy that stuff anymore as making the solution is way cheaper and works 
just as well.  I want to say it was like a 1% acid solution in alcohol??  What 
was the acid?  For some reason my brain says glacial acetic...but time has made 
me forget.  Is the alcohol you mix it in 100% or something lower with a water 
content to it?  Please help my alzheimers =)

We make our own here too. I can't remember why exactly. One reason is the space

I don't suppose that anyone out in HistoPersonLand has done a cost
comparison between making your own 10% NBF and purchasing a 50-gallon
drum of 10% NBF, perhaps?  I did this several years ago but my idea of
saving tech time/costs by buying the bulk was shot down because there
was someone here that did the making.  Now we're even more shorthanded
and I think it would be a better use of everyone's time to just buy the
darned stuff in bulk.  Any chances that someone's already done this?  If
you have, I'd appreciate your input and my everlasting gratitude
(brownies not included).

That could also be tendonitis related to the snuff joint. It starts with a D. 
Dequarveins or something, someone will know. I got it when I was pregnant with 
my second child. There are exercises for strengthening that tendon.

 I bought a big countertop jar opener a few years back for the same reason, 
only we have very large jars that we can't even get our little female hands 
around. The jar opener was supposed to open all sizes, but it wasn't able to 
open either our large ones or our small ones. Big waste! I hope you find 
something that works. Maybe one of us should invent jars that are really 
airtight and still easy to open!

I am looking for diff-quick stain kits that are economical.  My pathologist is 
contemplating another medical mission trip to Haiti and I need to locate a kit 
that she can take with her for blood smears.  I have found a few upwards of 
$600 but she does not need a gallon of each of the solutions.  If you know of a 
supplier of smaller volumes or even if you have protocols where you make your 
own, please share.

Use fabric softener sheets. You can wipe the blades and knife holders with 
them. My daughter's American Girl magazine says to pour lotion in your hand, 
rub it between your hands and while it is still shiny run your hands through 
your hair to control static.

I am looking for a vendor of BVDV antibody for IHC testing. We had used 
Syracuse Bioanalytical in the past. They seem to be gone. I would appreciate

Hi Laurie,

I purchased them because I am thinking about taking the HTL in the next year or 
so.  The are pretty nice, they are divided up into categories (fixation, 
processing, etc) and each category has a different color edge on the card.  
They have questions on the front with the answers and explanations on the back. 
 They were pretty expensive (75-80, I think), but would probably help out a lot 
when studying for an exam (a non-histologist could ask the questions and help 
you study)!  I will probably enlist my 9 year old, who gets a kick out of this 
kind of thing.

[Histonet] IHC protocol for vimentin in animal tissue

Our lab recently had our CLIA inspection and the surveyor informed me that we 
need to discard any dry chemicals with an opened date of more than 10 years, 
even though none of the dry chemicals have an expiration date. This was not 
written up as a deficiency, but it was suggested we follow up on this for our 
next re-certification.
 I am sure it is best to keep the chemicals current, but is this what all labs 
are doing?

It is not just Dako, but many vendors have jacked up the cost of goods.
Let's take the PSLIM from Accuplace. About a month ago you could
purchase this for $5K. Now that Thermo has acquired distribution rights
it costs over $10K and nothing has changed. To me, it is better to sell
10 at $5K than 5 at $10K. We were ready to purchase several more of
these, but have to reconsider our options now. No wonder Healthcare
costs are out of control.
