 This is a small research lab in the College of Veterinary Medicine. There

[Histonet] re-processing help

Hello Histonet,

We have issues- please advise! Long story short- some tissues did not get 
processed over the weekend, they went through the process but were not 
submerged in ANY (we think) of the reagents. Now they are shriveled up and 
brittle (they are intestinal). Is there any way to save them? Can we re-process 
them? Has anyone ever had this problem and had success recovering?

Hello Histoland,


A researcher brought us a group of poultry feather shafts to embed and section 
for routine and IHC staining. Has anyone out there had any experience with this 
that could share some advice on getting some successful sections?


So far we have tried facing the blocks and then applying "Nair for Men" for ten 
minutes to the block surface, followed by soaking the blocks for some time 
before attempting sectioning but still we have not had any luck. The sections 
turn out with large holes and blow apart on the water bath.


Any tips would be appreciated, thanks!

[Histonet] Leica embedding station

Hello all in Histoland!

We have a Leica EG 1150 H purchased in 2002 and we are starting to have issues 
with it not heating in the cassette and mold warmers as hot as it used to. The 
settings are all the way up to the max (70) but when I measured the actual 
temps it is in the low 50's. Has anyone else had this issue and if so would you 
mind sharing how you dealt with it?

[Histonet] microarray question

Hello Histoworld,

I was wondering if anyone out there has any experience with microarrays? Making 
the blocks and cutting them. We have been having a hard time with all the 
nuances of this process and were wondering if you could share any tips/tricks 
you have learned doing this process. Recently, I have been wondering if there 
is a special kind of embedding medium that may work better. We have tried with 
Paraplast and Paraplast Plus and it seems like regular is better in general. 
But even still, we have cracks that form, and sometimes the block pops off 
while cutting on the microtome and also we never seem to have every punch 
unroll when laying the ribbon out on the waterbath.

[Histonet] looking for diff cell counter

Good Morning Histonet colleagues,

My PI wants me to purchase a differential cell counter and I was wondering if 
anyone had any recommendations for a basic manual counter. One that offers the 
ability to count 5 different cell types and dings at every 100 cells. I found 
one through Fisher but it seems pricey. Does anyone know other places to get 
them?

Hello HistoWorld,

My lab is desperately trying to find where to purchase "Cranberry Aqua Source 
Nitrile Gloves" and having no luck. Going to the website listed on the box of 
gloves, there is no way to purchase the gloves online. It just says 'to order 
contact your local dealer'. I called their 800 number and they said that you 
have to contact a distributer and they recommended Henry Schien. When I 
contacted Henry Schien they were very snobbish on the phone and said that there 
was no way to get a quote from them if we aren't already "members" and account 
holders with Henry Schien. Just thought I would ask fellow histotechs if they 
use these gloves and if so, who their distributer is.

[Histonet] plastic section gram stain

Hello histonet world,

My pathologist just gave me a slide with a section on it that is intended for 
electron microscopy. Has if anyone ever done a Brown and Hopps special on a 
section embedded in Epon/Araldite?  She told me that her colleague has had the 
sections stained successfully with T-Blue on a hot plate before. But gram 
stains are a bit more complicated than a T-Blue! I can heat the staining 
components (i.e. crystal violet, safranin o, and iodine) but there are removal 
steps that require use of acetone, formaldehyde, acetic acid, and picric acid, 
and I am not sure how those reagents will work with the plastic. Any help or 
experience to share would be wonderful.

Blades for grossing tissues- anyone have recommendations? The vendor that we 
were purchasing just straight razor blades from has stopped manufacturing them. 
We don't use scalpels because of their higher cost and we are only working with 
poultry samples.

[Histonet] processor died overnight

I came in this morning to find that the processor died halfway through process 
last night. The tissues are in 100% ETOH exactly half point. We do have a back- 
up processor. In your professional experiences, would these tissues be 
salvageable? Could I create a new program on the backup processor that finishes 
the process from that point and transfer the tissues over?

[Histonet] IHC question

Hello Histonet world,

Our lab has an old Dako Autostainer for IHC and there is a leak in the y-axis 
arm at the point where the tubing goes through a juncture. I was wondering if 
there is anyone out there that uses this in there lab? If so, where do you get 
spare parts for it? Our PI wants us to try to replace the tubing ourselves to 
see if that fixes the problem before getting service.

Our pathologist wants us to do Sheehan's modification Giemsa and we don't have

Hello all in Histo Land,

I was wondering if anyone out there works in a Histo Lab that shuts down for 
the holidays? We are closed the week of Christmas and I am preparing the 
laboratory for sitting unattended for a week. If anyone else is in the same 
boat, do you have any tips for what you do to shut down the lab? We are 
planning on emptying the strainers and keeping the coverslipper nozzle 
submerged in xylene as air-tight as possible. Not sure what or if we should 
bother the processors...

mean, how many points they are out of? It says what your total score is out of 
400 but then the subtest scores are all high numbers that do not add up to 400, 
they add up to more so how do you interpret them?

[Histonet] bone saw

Hello histoworld,

Does anyone out there use a bone saw in their lab? We routinely have research 
cases with hundreds of femur head submissions from avian species. We currently 
use a bone saw made by Buehler from the 70's or 80's and it's a work horse, but 
the blade keeps cracking in the diamond tip from overuse during these surveys 
of hundreds of bones. I was wondering what kind of saws are out there that 
could be used for this purpose and if anyone has any experience with this? I am 
looking for something a little more durable, or if not, at least a little 
cheaper. Each blade costs about $350.

[Histonet] siphon drum pump

have three and they are all leaking at the squeeze/bulb area from the xylene

We got a case submission of a whole 44 week old chicken head and the write up 
states "swollen head and dirty ear canals". This is the first time we have seen 
this, even the pathologist is stumped. What part(s) of the head would you 
gross, and how?

[Histonet] bone saw blades

Hi all, in our lab we have a Buehler Isomet Low Speed Saw that looks like it is 
from the 80's. Up until about 7 months ago, we have used the bone saw fairly 
infrequently. As far as I know, this bone saw wafering blade has never been 
replaced. We now have a researcher who regularly needs sections of bone cut and 
the blade is getting much more frequent use, like hundreds of bones per 
project. My question to you is, how long do these blades stay sharp? Do they 
ever need sharpening? What is their lifespan? What kind of maintenance do they 
need?

from Biogenex by breaking the tablet in half and using half the amount of
