Hello All,

Our EM lab melted a block meant for IHC staining and put it into plastic. Is 
there any way to get the tissue back to paraffin?

I know it's a long shot but just wondering

[Histonet] Staples and Sutures

Hello All,

Recently we have had a marked increase in staples and sutures in our tissue 
that arrives for processing. I have tried to relay to our pathologists and PA's 
the difficulty in cutting sections and the tissue destruction in getting 
staples and sutures out once the tissue is processed to paraffin. I was told by 
some of them that in their experience histology should dealing with this issue. 
Though my own opinion is this is position is not valid, I've been asked to 
collect opinions from other histology departments if there are protocols that 
the grossing individual should be removing the foreign object. Please give me 
any feedback...Thanks

You will also see the cornflaking if your tissue is lifting off of the slide at 
all.  We used to get this more often on hard, decal specimens than on other 
specimens.  We used the film to coverslip.  If you remove the film from the 
problem slides and recoverslip conventionally with extra mountant and glass 
coverslips, I'm sure you will not see the artifact.

I am looking for help with cornflaking (tiny, brown dry spots under 
coverslip)artifact.  We have been using fresh xylene on our stainer and 
coverslipper, cleaned and wiped all containers dry before filling, tried 
different lots of coverslipping film and had service on our coverslipper to 
make sure it was functioning properly, including the xylene drip.  We continue 
to have this artifact and it is driving us crazy.  It is sporadic with no 
pattern of tissue type or placement on the slide.  Sometimes it lands on tissue 
other times not.  Most of the time when we remove the coverslip and 
re-coverslip it goes away (I am assuming because the acetone removes any minute 
amounts of water that may be present).  We just cannot figure out where the 
water is coming from.  Has anyone seen this artifact while using the drying 
step on the prisma stainer?  We just recently started using the drying on some 
slides and I am thinking maybe it is causing humidity???  I cannot say for a 
fact that our cornflaking started at the same time, but it is suspicious.

[Histonet] Vacuole appearance in lymph Nodes

Hi All,

I have sent this question to respected colleagues individually so pardon my 
redundancy if you have this email already.

Lately we have had random nodes show up with areas that appear to have multiple 
vacuoles through-out the specimen.  They have different patterns section to 
section so I know it is not in the tissue. The staining looks fine and the 
tissue cuts well so I don't think it is a processing issue. Any ideas on the 
problem would be welcome. Thanks

[Histonet] Whole Mount Immunos

One of my pathologist would like to do PSA, P504S (alpha-methylacyl-CoA
racemase), and CD31 on whole mount prostates. Is there anyone out there
doing this and if so could I ask some questions of you? We have not done
immunos on whole mounts before and since no automation supports whole
mount slides (we have Ventana and Dako) I would have to go back to
manual...which I haven't done in a LONG time. Recommend any kits,
protocols, ect...Any help would be appreciated. Thanks,

[Histonet] Full Mount Immunos

Does anyone know of any companies producing charged full mount slides? I
have a pathologist that wants immunos done on full mount prostates and
the tissue keeps falling off. I have looked, but cannot find any charged
