   I have always wrapped the cell blocks in lens paper.  If it is a messy one I 
wrap it 2 or 3 times in lens paper.

What is everyone doing regarding cell block preparation?  We have been using 
sponges for processing but we've had a recent contamination issue that may be 
attributable to the sponges (fluid types vary but mostly pleural fluids).  We 
will be testing sponges, biopsy bags, and paper...
anyone have a method they prefer?

[Histonet] Pregnancy

Hi Gang,
   Thought I would ask some people with some more experience than I in this.  
One of our techs is pregnant-  Yay !!!  She is very early - about 5 weeks.  My 
Lab Director wants her to start wearing a mask due to the chemicals.  So we 
need to ask what sort of precautions need to take place during the pregnancy 
etc.  We also coverslip by hand here out of xylene etc.  I can not remember 
ever working with any pregnant HTs in the past so I honestly do not know.  
Please carbon copy replies to jascholefi...@skaggs.net as well.  We 
predominately work with 10% formalin, reagent grade alcohols, paraplast, 
xylenes, clear-rite 3, Decal I and II (from Surgipath), cytofixative and some 
routine special stain items. Basically the typical myriad of stuff a very small 
routine histo lab would have (no immunos)

Hello all,

I was wondering what most people use as the first reagent after the 
formalins on their tissue processor?  We have always used a sequence of 70%, 
80%, 95%, and 100% but is anyone using 80% or even 95% to start their 
dehydration?

We use start at 95%, but we have two Pen Fix (alcohol fixative) steps between 
our Formalin and alcohols.  10% NBF, 10% NBF, PenFix, PenFix and then the 
alcohols starting at 95%.  

Speaking of 10% NBF has anyone ever used un-buffered formalin for routine 
processing?  I heard of a place using un-buffered formalin and wondered if 1) 
would that be okay or is it harsh on tissue and 2) would it keep the salt 
deposits down?

Hello all,
 My lab is looking into xylene substitutes, and I'd love some feedback on what 
other labs are using. We currently use SubX, but are there other items out 
there more economical? 


We use Clear-Rite 3 made by Richard Allan and is purchased at Cardinal.  I do 
not think it is very expensive we buy in a case of 4 gallons.  It does a decent 
job for clearing in the processor and de-paraffinizing slides, but we do 
coverslip out of xylene at the end of the stain line.  Actually we never tried 
to coverslip out of it Clear-Rite 3 so can not honestly give you a critique in 
ref to that.  We keep both on hand. Xylene for machine cleaning and 
coverslipping and Clear-Rite 3 for processing and de-paraffinizing. I like it 
better than Americlear and ProPar.

[Histonet] Xylene/Paint Thinner

I have a colleague who is convinced paint thinner is exactly the same as 
the SubX we use. Does anyone know if this is true?
Haha- funny you should say that.  I always swore that Mitaban (for canine 
demodex mange) was made out of xylene.  Almost looks like watered down xylene.  
Well I checked the MSDS and it is but only the inactive ingredient part at a 
high %.  On a lighter note that orange fragrance d-limonene flea dip is 
basically the same stuff as the Americlear is.

[Histonet] Bronch Order Forms

 Hi All,
  Not sure how many of you do cytologies in your departments?  We do quite a 
bit of bronchs around here and about 5 years ago a new physician who is no 
longer here developed the form that we currently use.   It has never been liked 
by the nurses in scopes etc and it is pretty complicated.  It allows Drs to put 
up to 5 microbiology, hematology, cyto or histo samples (many of which we share 
in all depts) on one form and has multiple check boxes to check as to what 
tests they want for each sample in multiple columns.  We are looking to 
streamline this form to hopefully make it easier for everyone involved.  Any of 
you have any Bronch ordering forms you really like or have used some in the 
past. Needs to be a paper order as we are not on a Lab system in Path at this 
point.

[Histonet] Iron Stain

Hi All,
  Is there any specific reason why one iron stain kit I have recommends that 
you air-dry the bone marrow and blood smears after the Fe stain is completed 
and then coverslip them while other kits don't mention that at all.  I have 
always stained my smears then dehydrate, clear and coverslip them like the 
tissue slides (at the end). Ergo Fe stain, rinse, paragon, rinse, 95% Alc, 100% 
Alc, xylene and coverslip.

[Histonet] AFIP Closing

 Sad that is where I went thru Histology Training at the Tri-Service School of 
Histotechnology in 1985.  I was in a class of 10 Air Force guys and I was Navy 
and there was one other Navy guy.  I was the only female in my class.  It was 
interesting and have been doing Histology non-stop since then- wow 25+ years 
has gone by fast.

[Histonet] Dictaphone System - HELP !!

Hi All,
  Does anybody use the old Dictaphones ?  We have been using them and the 
company does not support them any longer.  The one at our grossing station went 
down Friday and was replaced by a newer one (as we had purchased several from a 
hospital that was getting out of them).  This one did not have a foot pedal so 
a foot pedal connection was adapted to it; however, when the other machine went 
down it was unplugged by another team member and all the programming was 
therefore lost.  Does anyone have Dictaphones that are operated by means of a 
foot pedal who's IT team would be willing to share the proper method to program 
in a foot pedal to use w/ it.  It is almost impossible to gross without a foot 
pedal. Thanks in advance.

[Histonet] What New Tissue Processor Should we get ?

Hi All,
  If given the choice- what would be the best Tissue Processor to get for a 
small Path Lab.  We have always had VIPs. Ours is about 16 y/o and we need to 
replace it and use it as a back up or for small biopsies. I was looking at the 
new VIPs and our medical repair people are very happy w/ the VIPs in the past.  
I am looking at VIP 5 and 6.  Do not need anything too fancy.  Just want a 
workhorse that has little or no issues and is user friendly. 

Any input is much appreciate on these or other products - private emails great !
