[Histonet] GMS Question
Hello, I think I already know the answer but I'm not sure why so if someone can help me understand the theory behind it, I would greatly appreciate it. Currently, we use the Richard Allen kit for the GMS stain and it uses Periodic Acid as the 1st step. We use a control tissue from a case we had that was positive for fungus and it's a fungus ball from the Rt Maxillary. We ran a test for fungus on a different and current case of the same tissue (different patient): Rt Maxillary sinus. The control tissue did work, but the patient's tissue did not, so the doctor ordered a PAS for fungus and this clearly showed the fungal elements nicely. My question is why would the control and patient tissue have different results when they are both fungus balls from the same specimen source? Hello, We are considering a switch to a different brand of paraffin and this is because I feel we are having too many compressions in some of our tissue sections. Currently, we use Tissue Path Paraplast, regular. I would like to get feedback from you as to what you prefer. [Histonet] Her2 Fixation Requirement
Hello histoland I was just given the task to find a solution that is easy and will also comply with the CAP guideline for formalin fixation documentation, and so I started my research on the Histonet archives. I found some good information, but was hoping to get more feedback. Would you mind sharing with me the actions you are taking to comply with the guideline? We are a private lab and we provide histology/pathology service for 2 hospitals and a few surgery centers. We send our blocks to Genzyme for Her2, and we must document on their test order sheet how many hours the tissues have been fixed in formalin. I'm assuming I will need to start keeping a log here, with documentation that shows what time the tissue was excised and placed in formalin from the OR, and then documentation that shows the time it was dissected and then placed in the tissue processor. The problem that I may come across is getting the OR nurse to document the time for us. I don't know...maybe we need to put another sections on our requisition form, or maybe something on the formalin container itself for the nurse to write on. It'll be a hassle at first but if I can get the hospitals lab director involved, I'm sure it will work itself out. Anyway, if you wouldn't mind sharing some of your ideas, I would really appreciate it. Hello all - Is there a more effective and faster way to remove the film from the slide? I had to decolorize a couple of HE slides to do special stains, and it took 5 days for the film to remove. The slides were only a few days old and I soaked it in fresh xylene. I almost removed it too soon because the section was starting to lift off with the film. Proper plumbing, ventilation and lab space are serious considerations and fall by the wayside when these operations are put together. ~ Qualified personnel - All of us posting on this list understand the fact that qualified, competent Histology Lab personnel are in demand and difficult to find. The wage for qualified, competent technical expertise does not fit into the plans of the originators of these operations. ~ Proper equipment and instruments - While the aim of these operations is limited in scope, basic functional equipment and instruments are required. In the overall scheme of things, pathology is still a bargain. However, money must be spent to properly equip a competent working lab with modern instruments. I could go on but that makes the basic point. And Timothy, lest you think that I'm shooting from the hip I assure I am not. Like most people on this list, I am a serious, diligent professional. I have interviewed people from pod labs that have worked under horrible conditions. I am also aware of a pod lab trying to get off the ground that shouldn't have an ice cube's chance in hell. The plans for this lab do not have proper plumbing, ventilation or space and there is no equipment. While a position has been advertised I cannot think of a single tech, worth his or her salt, that would consider associating themselves with such an ill-conceived venture. One last point - don't forget Timothy, that one day you may be that patient. Or it may be your mother, sister, daughter, wife, son, friend...I'll stop there. Every block, every slide, every piece of tissue that I or any of my staff encounter is a precious patient...a human life. So trying to dumb everything down, cut corners and make a small number of people wealthy in the process is an irresponsible and utterly ridiculous risk. The professionals in our field have worked hard for a good number of years to come out of the basement spaces and after-thought little nooks and crannies and other poor facilities to do the highly skilled work this profession demands. I for one will never abide going backwards and I don't believe people of good conscience and humanity will either. [Histonet] Auto-IHC Staining Issues
Hello, I've been having a problem with the staining quality from an auto-IHC stainer and I was wondering if any of you experience the same thing. I notice sometimes that the staining in the tissue isn't complete, like there are areas where the fluid (one or more of the steps involved) didn't get in contact with the tissue, thus resulting in patchy, skipped staining. I'm thinking that this must be a dispensing problem. Is this a general thing with all auto stainers - ones like DAKO or Lab Vision's stainer or Biocare Medical's stainer? These stainers dispense fluid onto the slide. I hope this explains things and I really hope that I can get some feedback. It's getting very frustrating.
