[Histonet] Celestine Blue for Technovit 7200

Hi everyone,
 
I'm searching for a procedure or help w/a stain for thick ground sections using 
Technovit 7200.  The stain in question is a celestine blue/alum hematoxylin w/a 
Van Gieson counterstain.  Anyone have a procedure or recommendations?

Thanks Jack, what's the difference between the celestine blue w/alum 
hematoxylin and the Sanderson's?  What's the difference between the Sanderson's 
and the Masson's Goldner's?  Have you tried the celestine blue stain?

Why not use Sanderson's Rapid Bone Stain with Van Gieson picrofuchsin? You can 
get the Sanderson's and one or all of three counterstains for this particular 
kind of staining in a kit from Dorn and Hart Microedge (www.dornandhart.com 
http://www.dornandhart.com/ ).

[Histonet] RE: Cell phones

The cell phone do not call list is real.  In Ohio several people I know started 
getting lots of 'spam' calls on their cell phones.  They called their carrier 
(verizon) and asked how to stop it.  They recommended the government do not 
call list.  Once they called that number the spamming stopped.  If you read the 
snopes website the information stops in 2006.  Call your cell phone carrier if 
you are concerned instead of checking the internet if you are receiving these 
annoying calls.

I also looked at the Surgipath image, my slide was much more brilliant then 
that one.  I did use the acid fuchsin counter stain.  I just wasn't sure if 
mine would be considered 'over stained.'  I'll send you an image that Jack 
Ratliff sent me that compares to what I was seeing today.  I'm not using GMA, 
but Technovit 7200 ~100 micron thick ground sections.

On looking at the Surgipath photo, the bone is very pale although the
osteocytes, osteoid and soft tissues look good.  I am not sure what
counterstain (two are touted in the stain data sheet) was used as seen in
this photo.   One is a basic fuchsin that turns the bone red.  Be careful of
overstaining with fuchsin counterstain.  The other is light or fast green
and since we never used that, I can't comment on the color contrast with
bone and soft tissue components.  This may be the stain used in
photograph???



It always is a good idea to look at the stain while performing the actual
staining method.  After prescribed or desired time in RBS, simply rinse
quickly with running hot tap water,  blot and examine under the microscope
to see depth of stain for soft tissue/bone components. To view a dry
section, simply place a coverglass on top of section for higher power
examination.   If components stained by RBS not dark enough, you can return
to the hot RBS.  Rinse then blot, and counterstain after that.  It does not
hurt the bone section to dry.  In fact we preferred a dry section when going
back into either stain just to prevent dilution of the stain solution,
particularly when doing hand ground thick section surface staining.

Here are my responses:
 
 
Suzanne:  I no longer use Sanderson's, however we did have nice results on 
plastic embedded sections, in conjuction with other calcified stains. Use 
caution in over-staining. I do not recall ever reading about usage on paraffin.
 The Villanueva Osteochrome bone stain can also be used on calcified - we used 
it with fluoro labels, and then viewed using confocal microscopy with beautiful 
architecture and active new bone formation.  Vicki K

 

Sanderson's - we've not used it on wax, but we do use it on 15-20 µm ground 
sections. 

Method for resin:

*   Pre-heat Sanderson's to 55-60°C. Stain sections for approx 20 minutes 
(check every 5 minutes). 
*   Wash slides in warm water and dry with tissue 
*   Counterstain with Acid Fuchsin solution for 1 ½ minutues. (2.5g acid 
fuchsin, 2.5ml glacial acetic acid, 500ml distilled water) 
*   Drain slides and dry with tissue. 

 

I use this stain two ways routinely for thick or ground sections (35-50 
microns) embedded in MMA  DBP. You can stain with SRBS alone @ RT for 3-5 
minutes after a brief acid etch in 0.7% FA for 30 sec (rinse in DI water and 
blot dry) or combine it with a Van Gieson picrofuchsin counterstain for 1-2 
minutes (rinse in 100% EtOH and blot dry) after staining first in SRBS @ 60C 
for 10 minutes (rinse in DI water @ 60C and blot dry).
 
I have never used or needed to use SRBS on thin plastic sections because I 
routinely use MacNeals tetrachrome combined with Von Kossa and a Modified 
Goldner's to obtain all my bone related information. With that said, I have 
also never used this stain on paraffin sections and would be interested in the 
results if you try it.

look like Toluidine blue-- all shades of blue  nothing else.  Now I just stick 
with von kossa and/or tetrachrome for those.

[Histonet] Staining racks

Hello, I work in plastics using Technovit 7200.  We use plastic slides 
measuring 2x4cm.  Does anyone else use slides this large and if so, what do you 
use as a staining rack or slide holder?  They are too big to fit in the 
oversized glass slide racks.

[Histonet] Sanderson's Rapid Bone stain

Hello, I posted earlier regarding the Sanderson's Rapid Bone stain.  I forgot 
to mention that I will be using this stain on 75-100 micron thick ground 
sections  embedded in Technovit 7200 plastic.  
 
I am also wondering about paraffin staining using this stain.
 
Here's one response so far:
I've used Sanderson's on plastic sections a lot in the past, works well.  
Recently I tried it on rat tibias, decaled, FFPE.  Very disappointing.  They 
look like Toluidine blue-- all shades of blue  nothing else.  Now I just stick 
with von kossa and/or tetrachrome for those.

The tissue in plastic will be rabbit femurs I believe.  We haven't started 
anything yet, just getting references.  As to the paraffin staining, I believe 
that was an overall question from my director to see if she could incorporate 
that stain in paraffin as well as our Technovit 7200 work.
