[Histonet] Interfacing with Meditech Magic

Hello Everyone:
I am looking for some suggestions. We use Meditech Magic and we are trying to 
interface with the Leica Cerebro system. We are trying to put the correct info 
in the .hist section of Meditech so that Cerebro can pull the necessary info 
for the labels.
My unsolved issue at the moment is how to tell cerebro that I need an HE label 
and  special stain label on the same level. My computer person is saying that 
the second procedure typed in will over ride the first one ordered.

[Histonet] Alcian Blue

Hi Everyone:
Can anyone tell me why some Alcian blue procedures say 30 min in Alcian Blue 
and some say 10 min?Our control works well at 10 minutes but today I had a Dr. 
say that he expected a small amount of cells to stain and they didn't in an 
esophagus bx.So, now I'm wondering if b/c most of the procedures that I've read 
say 30 min, that's what would be best?

We noticed for any tissues that have a tendency to float off:
Leave over night at room temp to air dryThe next day heat in a separate oven at 
56 degrees for 15minAllow the slides to cool back down to room temp.Then stain 
without using the oven on the stainer.Works great for fatty tissues
Of course this is only great if you have a day to spare. :)

One of my pathologists wants me to look into Osmium Tetroxide for staining 
lipids. From what I can gather on the internet, it looks like it is used in 
Electron microscopy for fixation and staining.
Is anyone using this procedure for routine 4 micrometer sections?

 what does everyone currently do with the vials the Pathologists empty when
 doing gross?  Ours goes in biohazard bags, but someone mentioned to me they
 the labels should be taken off and then the vials put in the biohazard bag.
 If we did this we would just be taking off labels all day long. I have
 worked at 2 hospitals in my 35 years of experience and we have never done
 this.  If we started this look at the clinical labs with blood specimens.
 Isn't there some sort of confidential agreement between the hospitals and

[Histonet] Number of blocks before a processor change???

We currently change our processors every 5 runs. 
 
I am thinking it would be more cost and time effective to count the number of 
blocks before doing a change. If you count your blocks could you share with me 
at what number you do a change. We run all types of tissues.
We use a gallon of reagent in each position. Vip5.

[Histonet] prostate bx containers

Has anyone used these prostate biopsy collection containers from Simport? It 
looks like a petri dish and has 12 individual wells in it, each one labelled 
with the location from the prostate. 
We are going to trial them but I would like some opinions on them. 
We currently get 10 to 12 bottles per case. So, all the bx's could go in this 
one container.

I have a 60 yr old awesome tech in the grossing room. She is wonderful and is a 
great teacher for the new younger techs. She works straight 9-5 everyday. I 
value her experience and want to keep her work environment as accomodating as 
possible. I also have a wonderful lady that only works in the microtomy room, 
again same theory with keeping her happy. 
The new younger ones do rotate every bench. I see how unhappy the older techs 
in core lab are b/c they have to work equal nights with the newbies. 
I believe there should be some appreciation for the ones that have been there 
for 30 plus yrs. 
Techs are waiting in line to work in our dept. 
I also love my job b/c the ppl I work with are so great. :)

 rotations and jobs no matter the age. It is sometimes difficult due to some 
 internal changes in the locations of the Gross Room and Histology (literally 
 a quarter mile apart) however; we all work with what we have in these areas. 
 Occasionally someone will object to doing a job and it is explained we all do 
 everything as we are short handed and have no choice.

I've never worked with goat knees, but we were experiencing lots of PIN4 
prostate biopsies floating off during IHC staining with HEIR.
Since we started cutting the prostate bx's at 3 micons, huge difference. They 
almost never float off. :)

 Your test subject is one of the more difficult there are.
 Although you may get some background, use some gelatin dissolved in the water 
 bath to try to adhere the sections to the slides. Also try to cut as thin as 
 you can and make sure there is no water left between the sections and the 
 slide.
 Bake at 60ºC for at least 45 minutes before starting the IHC procedure. If 
 you use HIER try to switch to pepsin.
 The least stress you add to the sections the better.

 are decalcified FFPE sections and we are using a Dako autostainer. We are
 currently trying air-drying for  1-2 days  and baking of tissue slides
 before use in IHC..
 
 The one previous time we tried baking, we noticed a decrease in IHC signal.
 The sections are pretty large, we tried cutting them down, but can't
 decrease the size much.  We also  decreased the number of washes and size of
 washes on the Daok, which helped only slightly.  Does anyone have any

We were having issues with placenta contamination aswell. Our Dr.s now wrap the 
placenta sections in tissue paper and this seems to have cut down on the 
problem.

We are short histo techs and are using aids to help more and more. Our manager 
believes they can be doing the grossing of a histo tech as long as they have a 
tech or Dr to consult with. Can someone remind me of the guide lines for 
grossing. One has a BSc, is that enough?
